چکیده

Sir: Phenomycin, a basic polypeptide antibiotic, exhibits significant inhibition of Ehrlich carcinoma, sarcoma 180, and adenocarcinoma 755 in mice but has no antimicrobial activity1'2). The mechanism of action of phenomycin has been studied3). The antibiotic inhibits protein synthesis to a significant extent in mammalian ribosomal systems, but much less in bacterial systems. The selective toxicity of the antibiotic is similar to that of diphtheria toxin. Moreover, both are simple proteins. The activity of phenomycin on protein synthesis was comparatively studied with diphtheria toxin. It has been observed that diphtheria toxin inhibits protein synthesis only in the presence of NAD (nicotinamide adenine dinucleotide) and that transferase II is blocked by toxin and NAD4>5>6). The effects of phenomycin

Design and Production of Recombinant TAT Protein Structure, Catalytic Domain of Diphtheria Toxin, and Evaluation of Its Effect on Cell Line

Background and Objectives: Cancer is one of the most deadly diseases in the present age and its conventional therapies have had low success. Toxin therapy of cancer is a new therapeutic approach, which has attracted the attention of pharmaceutical specialists. Diphtheria toxin consists of three functional, transducing, and binding domains, that the functional part inhibits protein synthesis and...

Mechanism of action of phenomycin, a tumor-inhibitory polypeptide.

studied, using mammalian systems. Phenomycin was observed to inhibit protein synthesis, but not RNA or DNA synthesis in growing HeLa cells. Approximately 50 % inhibition of leucine incorporation into protein was demonstrated at a concentration of 100 mcg/ml of phenomycin when the cells were treated with the antibiotic for 24 hours. Phenomycin significantly inhibited protein synthesis in the lys...

Short Communication: Diphtheria Toxin Repressor (dtxR) Gene-based Genetic Diversity of Corynebacterium diphtheriae isolated in Jakarta, Indonesia, 2018-2019

Background and Objective: In Indonesia, diphtheria cases caused by Corynebacterium diphtheriae are still occurring until today. One of the causes is probably the diphtheria toxin repressor (dtxR) gene which influences toxin expression. Therefore, in this study the characterization of the gene was performed to determine the mutations that affect the DtxR protein.   Methods: The dtxR genes of ...
